Thyroxine is produced in the thyroid gland of all mammals.
It is found in useful quantities in the tissue of the thyroid gland.
Unlike insulin which is broken down by stomach acid and very poorly absorbed, thyroxine is not broken down in the stomach and is absorbed.
The amount you need is trial and error - there is no accurate way
You would judge the dose based on history and physical findings
Not enough - feel sluggish, mentally slow, tired constantly, weight gain, dry skin, slow heart rate, low body temperature
Too much - weight loss, flushes, fast heart rate, anxiety, high blood pressure, slightly raised resting temp
Most people who are diagnosed with hypothyroidism will know what they felt like before they were diagnosed and how it changed when treated - this will give a ball park idea of if they are getting enough or to much thyroxine.
Pigs (followed by sheep or cattle) are probably the best option to source thyroid tissue - but in theory any mammal will do. It is likely concentration may vary from batch to batch so ongoing dosing will be hit and miss.
There are two options for preparing animal thyroid gland:
Drying - the thyroid gland is sliced really thin and air dried or dehydrated. Dosing can occur by eating the dried slices or by pounding the slices into powder and taking a measured amount
Extraction into alcohol solution - the thyroid tissue can be macerated and mixed and allowed to sit in an alcohol solution - this is them filtered to remove any particulate material. The remaining tincture will contain an amount of active thyroxine. The exact shelf life I don think has ever been determined. But it should be effective for a month or so - your mileage may vary hugely here. I found one reference that commented that alcohol broke down thyroxine, but this was a single reference and there are numerous older references to this being an effective way of extracting thyroxine.
